Testing Longevity in Mice
Researchers recently completed a study on semaglutide, a GLP-1 medication marketed as Wegovy and Ozempic, that indicates the drug may extend lifespan. Scientists administered the medication to female mice starting at 20 months old. In human terms, this age matches a person in their 60s. The team monitored these mice until the end of their natural lives. When compared to a control group that received only saline injections, the treated mice lived longer. Their median lifespan increased by roughly 12 percent, moving from 742 days to 834 days.
This research suggests that semaglutide might offer benefits beyond blood sugar management and weight loss. The mice treated with the drug exhibited better physical performance than their untreated peers. They balanced longer on rotating rods and navigated mazes with higher success rates. These improvements remained visible even after researchers adjusted the data to account for the leaner body mass of the treated mice. The treated animals also showed less tissue inflammation.
Links to Calorie Restriction
For two decades, Danica Chen has studied calorie restriction at the University of California, Berkeley. She suspected that semaglutide might act as a proxy for this type of dietary intervention. Calorie restriction has long been associated with extended longevity in laboratory settings. Because GLP-1 drugs suppress appetite, Chen hypothesized that they might replicate the internal effects of eating less without requiring the strict willpower typically needed for a low-calorie diet.
To verify this link, the team conducted a follow-up experiment lasting five months. One group received semaglutide while the other group simply consumed 24 percent less food. This reduction matched the caloric intake of the drug-treated mice. Both groups showed similar gains in muscle strength and coordination. However, the semaglutide group performed better on tests measuring memory and curiosity. This suggests that while the drug mirrors the physical benefits of dieting, it carries unique advantages.
Genetic Pathways and Future Questions
The researchers sequenced gene activity in the livers of the mice to understand the underlying mechanics. They compared this data against established patterns for calorie restriction. Both interventions quieted inflammatory responses and altered fat processing. They also activated specific genes responsible for blood sugar regulation and protein maintenance. Still, the drug provided better metabolic control and cognitive health than the diet alone.
Questions remain regarding whether the drug acts as a perfect substitute for traditional dieting or if it serves as a separate, more potent intervention. Nir Barzilai, a longevity researcher at the Albert Einstein College of Medicine, noted that the current data does not confirm whether a restricted diet might ultimately provide a longer life than the drug. Chen confirmed that her team is continuing to investigate these comparisons. Her group is currently running tests to see how these results translate into longer-term health outcomes. The field of longevity research continues to look for evidence that these pharmaceutical interventions can safely transition from laboratory mice to human health applications.
