A new study co-led by Yale researchers reveals that a person’s genetic background does more than dictate cancer risk. It also dictates how tumors behave and change over time. Published in the journal Nature, this research indicates that inherited DNA influences which mutations a tumor acquires, its growth rate, and its level of genetic instability.

To isolate the influence of inherited genetics, researchers studied liver tumors in four genetically distinct strains of mice exposed to the same chemical agent. Despite identical exposure and environment, the tumors followed different developmental paths based on the host's genetic background. This confirms that the same mutation can trigger opposite effects depending on the individual host.

Lead author Sarah Aitken notes that inherited genomes interact with acquired mutations to alter critical cancer pathways. This discovery explains why cancers with similar mutation profiles often result in different patient outcomes. It suggests that current medical approaches, which rely primarily on age or environmental factors for screening, overlook a key component.

Moving forward, these findings advocate for integrating individual genetic profiles into diagnostic tools and treatment design. While researchers cannot yet pinpoint the exact genetic differences responsible, the study highlights a clear need to adjust screening protocols to account for inherited factors. This shift could improve how doctors predict clinical outcomes and design personalized strategies for patients.
