Recent research from the Psychiatric Genomics Consortium reveals significant genetic differences between two major types of depression. Researchers studied over 460,000 individuals to categorize major depressive disorder based on energy-related symptoms. The study separated patients into two distinct groups, AERS+ and AERS−, based on their physiological responses to the condition.
The AERS+ group experiences hypersomnia, increased appetite, and weight gain. In contrast, the AERS− group is marked by insomnia, decreased appetite, and weight loss. These groups showed different genetic markers across 27 distinct loci identified during the study. This research suggests that depression is not a uniform condition but rather a set of disorders with different biological roots.
The findings point toward metabolic health as a key differentiator. Individuals in the AERS+ group share genetic links with metabolic syndrome, type 2 diabetes, and higher levels of C-reactive protein. This group also showed higher SNP-based heritability but lower polygenicity, meaning fewer genetic variants often exert a larger effect on their specific symptom profile.
The AERS− group showed a different path. These patients displayed genetic correlations with anorexia nervosa and potential mechanisms linked to schizophrenia risk. While these results show distinct genetic architectures, researchers emphasize that they are not fully separate conditions and still share moderate genetic overlap.
This work aims to move psychiatry toward more precise treatments. By understanding the metabolic and genetic signature of a patient, doctors might eventually match individuals to medications that target their specific symptoms. Although clinical practice is not ready for these changes today, this study provides a clear framework for future treatment-matching trials.
